Neural differentiation and neurotrophic factors expression in rat bone marrow stromal cell

Objective To investigate the neural differentiation potential and expression change of neurotrophic factors-glial cell line-derived neurotrophic factor (GDNF) and nerve growth factor (NGF) in rat bone marrow stromal cells (BMSC). Methods BMSC were harvested from male SD rats and cultured in DMEM supplemented with 20% fetal bovine serum, and characterized by flow cytometry. At passage 6, BMSC were pre-induced by bFGF (10 ng/ml) for 24 h, then induced with GDNF (10 ng/ml) in fetal gut condition medium (FGCM) for 10 d. The expressions of neuronal markers NSE (neural specific enolase), NF (neurofilament), and glial cell marker GFAP (glial fibrillary acidic protein) were detected by fluorescent immunohistochemistry. The expressions of mRNA of GDNF and NGF were detected by RT-PCR. Results The cultured BMSCs were positive for CD90(99.7%) and negative for CD45 on flow cytometry. After 10-day induction, a certain number of cells adopted neuron-like morphological changes and expressed NSE and NF, but no GFAP by fluorescent immunohistochemistry. BMSC expressed low level NGF and GDNF mRNA, while after induction, the expressions of NGF and GDNF mRNA were highly increased. Conclusion GDNF can not only induce neural differentiation of BMSC, but also increase the expression of NGF and GDNF significantly.
