Establishment of classical eyeblink conditioned reflex model in conscious and unfixed guinea pigs and study of related acquisition rule

Objective To establish a model of classical eyeblink conditioned reflex(EBCR) in conscious and unfixed guinea pigs and to analyze the related acquisition rule.MethodsWith the aid of metal electrode embedding technique and simple follow-up air puff stimulus facility,conscious and unfixed guinea pigs were trained to performed EBCR with tone as conditioned stimulus(CS) and air puff as unconditioned stimulus(US).The eyeblink behavior was evaluated through electromyography(EMG) of musculi oculi.ResultsThe eyeblink EMG signal of musculi oculi were successfully collected,and the classical eyeblink conditioning was obtained.The acquisition rate in the 4th,7th,and 10th day were 31%,61.3% and 82.7% respectively,and the maintain rate in the 15th and 20th day were 81.3% and 82.3%.ConclusionConscious and unfixed guinea pigs can be trained to obtain EBCR with the techniques of musculi oculi EMG signal recording and follow-up air puff stimulating method.It is an easy,convenient,feasible,and reliable method with high achievement ratio.It could be applied to study conditioned eyeblink behavior of animals with big body type,or animals uneasy to be fixed.
