Affinity chromatographic separation and identification of goat HSV- 1 specific transfer factor

The active components of herpes simplex virus type 1 (HSV-1) specific transfer factor (TFHSV-1) were separated from dialysates of goats lymphocytes immunized with the corresponding virus using the affinity chromatographic technique. The biological activity and purity of the transfer factor components were then evaluated by 51Cr labeled leukocyte adherence inhibition (51Cr LAI) assay and hydrophobic interaction high-performance liquid chromatography (HI-HPLC). It was thus shown that the separated TF components induced obviously the splenic cells of normal mice to produce LAI effect on HSV-1 antigen, but not on control antigens, indicating that the separated components has the antigen-specific activity of TFHSV-1. Analyzed by HI-HPLC, the active components appeared only two peaks in the chromatogram. It seems, therefore, that partially purified transfer factor active components were obtained from the dialysates, and the affinity chromatographic method on HSV-1 antigen absorbent employed here is well suited for the isolation of complex dialysates containing TF.
