Expression analysis of tumor suppressor gene NDRG2 in breast cancer tissues and cell lines

AIM: To analyze the expression level of candidate tumor suppressor gene NDRG2 in human breast cancer tissues and cell lines. METHODS: Four kinds of breast cancer cell lines and twenty one samples of breast cancer tissues with matched normal tissues were collected. The NDRG2 mRNA level was detected by semiquantitive RT PCR and RNA dot blot and the Ndrg2 protein level was analyzed by western blot and immunocytochemistry. RESULTS: The results of RT PCR and western blot detection indicated that five of the twenty one breast cancer tissues had low NDRG2 mRNA level and low protein level. Three of the four breast cancer cell lines had low NDRG2 mRNA level by RT PCR, and two of the four breast cancer cell lines had low Ndrg2 protein level by western blot and immunocytochemistry. CONCLUSION: NDRG2 has low expression in some breast cancer tissues and cell lines, which indicates that the low level NDRG2 may be engaged in the development of breast cancer tissues and cell lines.
