Study of protective effect of interleukin-10 on ischemia/reperfusion-induced lung injury following lung transplantation

AIM: To investigate the protective effects of Interleukin-10 (IL-10) on ischemia/reperfusion (I/R)-induced lung injury following rat lung transplantation and to explore its mechanisms. METHODS: Thirty-six SD rats were randomly divided into 3 groups with 12 rats in each group: sham operation group (control), lung transplantation group and IL-10 treatment group. IL-10 was administered to the recipient rats through intra-trachea way. The wet/dry ratio of lung, lung permeability index and neutrophil percentage in bronchoalveolar lavage fluid (BALF) were detected. The serum level of tumor necrosis factor α(TNFα)was detected by enzyme-linked immunosorbent assay (ELISA). RESULTS: The wet/dry ratio of lung, lung permeability index and neutrophils percentage in BALF of I/R group were (6.57±0.89) ,(23.49±5.13)×10 -4 and (49.84±6.53)%, significantly higher than those of the control group(P0.01). The administration of IL-10 to recipient rats before the operation markedly reduced the indexes above to (4.23±0.58), (15.96±3.04)×10 -4 and (20.33±4.26)%, respectively(P0.01). The serum level of TNFα(μg/L)increased to (0.86±0.11) after lung transplantation, markedly higher than that of control group, however, the IL-10 treatment could reduce its level significantly to [(0.39±0.05),P0.01]. CONCLUSION: IL-10 protects the lung function against I/R injury after transplantation, which is related to its inhibition of TNFα release and activation of neutrophils.
