Effect of continuous dynamic compressive stress on expression of cyclooxygenase mRNA and related signaling molecules in experimental fracture healing

AIM:To explore the effect of continuous dynamic pressure stress on cyclooxygenase(COX) mRNA expression and prostaglandin E2 (PGE2) and cAMP contents during experimental fracture healing. METHODS: Unilateral osteotomy of left humeral diaphysis was performed in 80 adult New Zealand rabbits. The humeral fracture was fixed with swan-like memory connector (SMC) in study group and with 4-hole dynamic compression plate (DCP) in control group. Samples from the fracture gaps were obtained at 3, 7, 14, 21, 28, 56 and 84 days after operation. The temporal expression patterns of COX mRNA during the repair process were examined through real-time RT-PCR. The contents of PGE2 and cAMP were detected through radioimmunity method and the process and quality of fracture healing were observed histologically. RESULTS: The relative levels of COX-2 mRNA as well as the contents of PGE2 and cAMP were elevated after fracture, and were significantly higher in study group than those in control group at 3 and 4 weeks postoperatively. Histological observation revealed that the mineralization of cartilage matrix, the endochondral bone formation and the callus remodelling took place earlier in study group. CONCLUSION: The persistent dynamic longitudinal pressure stress produced by SMC may contribute to endochondral bone formation of callus and accelerate fracture healing, which is considered to be closely related with the signaling pathway that has several critical components including COX-2, PGE2 and cAMP.
