SEQUENCE COMPARISON OF WILD COMMON CARP POPULATIONS BASED ON PARTIAL MITOCHONDRIAL DNA SEQUENCES

Nucleotide sequences of 16S rRNA, D-loop,and Cyt b gene of mitochondrial DNA were amplified using PCR techniques and analyzed from wild common carp (C. carpio) populations and Barbless carp (C. pellegrini)to detect their genetic variability and phylogenetic relationship. The results showed that, 221 nucleotide variable positions and 217 parsimony-informative sites were detected in analyzed 1492 bp sequence among the populations. With Barbless carp as the out group, phylogenetic relationships among populations were reconstructed and the divergence time was estimated. The phylogenetic trees constructed by MP and NJ method were of very similar topology. Xinjiang population was different from the other populations with farther genetic distance. The populations without Xinjiang population were similar and clustered together. The divergence time between Xinjiang population was estimated about 2.42-2.45 million years ago.
