Effects of jujube date polysaccharide on tumor necrosis factor production and its mRNA expression in peritoneal macrophages in mice

Objective To understand whether jujube date polysaccharide (JDP-N) affects tumor necrosis factor (TNF) secretion and its mRNA expression in mouse peritoneal macrophages Mφ. Methods Cell proliferation was determined by MTT method and TNF-α mRNA by semiquantified RT-PCR. Results JDP-N stimulated TNF production that reached the maximum at around 6 h, demonstrating similar effect to that of lipopolysaccharide on TNF production. Protein kinase C played a key role in mediating TNF production of Mf induced by JDP-N, in the process of which enhancement of TNF-a mRNA expression in Mφ was also observed. Conclusion Promotion of TNF production might be one of the mechanisms by which JDP-N enhances the immune function.
