Determination of amoxycillin and metronidazole in human plasma by ion-pairing HPLC

Objective:An ion-pairing liquid chromatography(IP-LC) assay of high-performance was developed to measure the plasma concentrations of amoxycillin and metronidazole simultaneously. Methods: ZORBAX ODS(4.6 mm × 150 mm,5 μm) column was applied.The mobile phase was acetonitrile-20 mmol/L potassium dihydrogen phosphate buffer(containing 2.0 mmol/L octanesulfonate sodium,pH 2.5)(28:72,(v/v).) The flow rate was 1.0 mL/min,the wavelength was set at 230 and 313 nm for amoxycillin and metronidazole,respectively.The injected volume was 50 μL.Protein in plasma samples was precipitated with acetonitrile. Results: The calibration curves for amoxycillin and metronidazole assay exhibited excellent linear relationship in the concentration range of 0.2～50.0 μg/mL with a correlation coefficient of 0.9990 and 0.9982,respectively.The within-day and inter-day precisions were not more than 8.8 %.The quantitation limits of the two drugs were 0.2 μg/mL. Conclusions: The method is simple,accurate and allows to be used as analytical method for routine plasma monitoring and pharmacokinetic studies of amoxycillin and metronidazole.
