Identification of Aspergillus niger EIM-6 with High-yield of Pectinase and Optimization of Its Submerged Fermentation Conditions by Response Surface Methodology

A highly pectinase-producing strain EIM-6 was isolated and identified as Aspergillus niger based on its phylogenetic analysis of ITS sequence.Through single-factor experiments,the optimum carbon and nitrogen sources for EIM-6 to produce pectinase in shake-flask cultures were obtained,subsequently some process factors under submerged fermentation conditions were evaluated and three factors demonstrated prominent effect on pectinase production,respectively by Plackett-Burrman design.The results showed that the optimum medium contained w(orange peel)4.09%,w(wheat bran)3.16%,w((NH4)2SO4)0.35%,w(K2HPO4)0.3%,w(CaCO3)0.24% and pH 6.As a result,Aspergillus niger EIM-6 showed a maximum pectinase activity 30 231 U/mL at 28 ℃,250 r/min for 80 h and inoculum size 107/mL.The pectinase yield was improved 2.07 fold compared to the original conditions.
