The Improvement of Orthotopic Left Pulmonary Allograft Model in Rats

Objective To establish rat orthotopic left lung allograft transplantation model by the improved "cuff-like" vessel and bronchial anastomosis technique. Methods Thirty-six male Sprague-Dawley rats were randomly allocated to donors and recipients. The donor right hilus was clipped for 5 mins(clipping test), then the blood samples were collected from carotid artery for arterial blood gas analysis as baseline. The graft's oxygenation function was also evaluated after 2-hour reperfusion. Dynamic compliances(Cdyn), resistances of airway(Raw) and peak inspiratory pressures(PIP) were monitored before the operation and after 2-hour reperfusion. Results After 2-hour reperfusion, Cdyn, Raw and PIP were worse than related baselines, but there weren't significant differences. The PaO_ 2 , OI, Qs/Qt, and A-aO_ 2 were significantly worse than related baseline values(P0.01). This improved model makes the operation easier than ever. Conclusion This simpler model may be used to study lung transplantation.
