The Pathological Changes of the Amygdala and the Myocardium after Focal Cerebral Ischemia-Reperfusion Injury in Rats

Objective To investigate the neurological deficit and the morphological change of brain tissue as well as the pathological change of the amygdala and the myocardium caused by focal cerebral ischemia reperfusion injury(CIRI) in rats and to discuss the possible mechanism.Methods The right middle cerebral artery in rats were occluded by inserting a thread through internal carotid artery for 2 h, and then reperfused for 24 h.Using neurologic score to record the neurological deficit,2,3,5-triphenyltetrazolium chloride(TTC) staining technique to measure the infarct size of brain,the AutoCAD image analytic software to calculate the degree of cerebral edema,H-E staining technique to investigate the pathological changes of the brain tissue,the amygdala and the myocardium and to count the density of pyramidal neuron on the corresponding encephalic region in the infarct side.Results After middle cerebral artery occlusion 2 h/24 h reperfusion,the neurological function was deficient and the infarct focus of brain and cerebral edema was showed on the injured side of brain tissue.The infarct size and cerebral edema were both 0 in the sham group,while were respectively(21.95±2.13)% and(33.42±11.30)% in the model group.The ischemic necrosis and edema were showed in the cortex and amygdala,and in which the density of pyramidal neuron was decreased.The count of pyramidal neuron in the cortex and amygdala were respectively(41.56±6.50) and(40.96±4.30) in the sham group, while were(10.08±2.67) and(8.56±2.25) in the model group.Both severe hyperemia and infiltration of inflammatory cell appeared in the myocardial cell.Conclusion After focal CIRI,the brain tissue was injured severely,and the conspicuous pathological changes were showed in the amygdala and the myocardium.
