Iufluence of peroxisome proliferator-activated receptor γ(PPARγ) ligands on angiogenesis in renal cell carcinoma

Objective To investigate the expression of vascular endothelial growth factor(VEGF) induced by peroxisome proliferator-activated receptor γ(PPARγ) in renal cell carcinoma(RCC)-derived cell lines.Methods RT-PCR was performed to determine the VEGF mRNA before and after treatment with 0 and 50 μmol/L TZDs(pioglitazone or troglitazone) for 48 h in two RCC-derived cell lines(786-O and A498).The mutative expressions of VEGF protein were detected by western blot analysis.Results Treatment with 50 μmol/L TZDs(for 48 h) decreased both the VEGF mRNA and protein expression in RCC-derived cells.Conclusion The results reveal that PPARγ ligands involve angiogenesis inhibition on RCC cells.PPARγ may be the candidates target for a novel approach to the treatment of RCC.
