Effects of berberine hydrochloride on alphar_1-adrenoceptor and voltage dependent calcium channel in vas-cular smooth muscle

Wang Wenya, Chen Kemin, Guan Yongyuan (Guangzhou Medical College, PLA. Guangzhou,510315)
In the endothelium-removed thoracic arotic rings of rats, vascontriction induced by phenyle-phrine (phe) was inhibited by berberine hydrochloride(Ber). And Ber shifted the logD-response curve for phe to the right parallelly, without changing maximal response. The pA2 value was 6. 68*0. 60(n=10). In the radioligand-receptor binding experiment, Ber and H-prazosin can bind competitively with the same sites of alphar-adrenoceptor in the rat cell membrance. Its ki value was 5. 84 μm. Ber shifted KC1 and CaCl2 cumulative dose-response curve rightward and decreased the maximal response. The pD2' were 3. 78±0. 19 and 4. 22±0. 14(n = 8) respectively.
