BIOLOGICAL BASIS OF RADIOLOGICAL PROTECTION

The development of radiobiological effects, as the basis of radiological protection, was overviewed in the first part of this paper. Three topics were involved: (1) the effects of low level (low dose rate) irradiation and its risk assessment; (2) the rapid developments of radiation carcinogenesis, including target molecule, single cell theory, multiple steps of development, oncogenes and oncogene suppressors and characteristics of radiation oncogenesis; and (3) hereditary effects with emphasis on multiple factoral diseases. In the second part, experimental studies in this laboratory were reviewed. The malignant transformation of different cells (SHE, rat tracheal epithelial and human tracheal epithelial cell) after exposure to α particles were studied as the experimental model. In the process of transformation, five biological aspects were investigated: (1) biological characteristics of transformed cells; (2) chromosome aberrations; (3) molecular alteration of oncogenes and oncogene suppressers and their regulation; (4) analysis of related novel genes were by means of differential display; and (5) detection of p53 gene mutation in human lung cancers by PCR SSCP. In these series of studies, some new findings were obtained. Twenty four cDNA fragments which are closely related to the transformation have been cloned and 13 of them are novel and was registered in GenBank. In a preliminary study, one new whole length gene with 3.5 kb was found and it is being studied further. (
