A study of analyzing DYF155S1 locus for forensic medicine

Objective To explore the application value of analyzing DYF155S1 locus,one of Y chromosome specific minisatellite,for forensic medicine.Methods Methods Fluorescent MVR-PCR was employed.The PCR products were analyzed by ABI 377 sequencer.Also,Amp-FLP was analyzed with the polyacrylamide gel electrophoresis followed by silver staining.ResultsThe results obtained from different tissues of one individual were identical.With 0.3 ng genomic DNA,genotype could be successfully analyzed by Fluorescent-MVR-PCR method.The DYF155S1 genotype of the male/female DNA mixture with male/female components in the ratio of 1∶100 could be detected with 110 ng DNA template.The mutation rate of DYF155S1 locus was found to be 0.9% among 336 meiosis(3/336).The genotype of DYF155S1 could also be obuained by analyzing the vaginal-seminal-stain taken from sexual assaults with these methods.But these methods could only distinguished males of mammals from the non-mammals.Conclusion Fluorescent-MVR-PCR and Amp-FLP methods for analyzing DYF155S1 locus were reliable and highly sensitive for forensic practice.
