Separation of Flavonoids by Supercritical Fluid Chromatography

The separation of flavonoids, which differs in the position and number of substi- tuted hydroxyl groups in their chemical structure, by Supercritical fluid chromatography us- ing ethanol and phosphoric acid-modified carbon dioxide was studied. The effects of the com- position of mobile phase, column type,pressure and temperature were investigated. It showed that the composition of mobile phase is the most pronounced factor influencing the separation and the addition of a very small amount of phosphoric acid to the mobile phase brings about the separation of all compounds with good peak shape. The type of packed columns also effected the separation, and the phenyl column was most suitable to separate the flavonoids. The retention times of the compounds decreased with increasing pressures and increased with the decreasing temperatures under the experimental conditions. The ex- tract of Ginkgo baloba leaves was analyzed by this chromatographic system, and the major flavonoids in it were separated efficiently.
