Enantiomeric Separation of Amino Acid Derivatives by Ion-pair Chromatography

The enantiomeric separation of N-3 ,5-dinitrtobenzoyl(DNB) of amino acid derivatives by ion-pair chromatography with a chiral stationary phase(CSP), derived from (S)-N-(2-naphthyl)-alanine has been examined. It was found that the increase in the concentration of a chiral ion-pair reagent caused significant increase in the capacity factors of both enantiomers of DNB-phenylglycine and DNB-leucine until a limiting value is reached, while the enantioselectivities of both analytes remained unchanged. A mobile phase of methanol-aqueous phosphate buffer was used. It was found that the concentration of methanol in the mobile phase dramatically affected the retention of the enantiomers. A minimum in enantioselectivity occurred at about 60% methanol. In addition, the separations of the enantiomers of the same analytes on a (R)-N-(2-naphthyl)-alanine-derived CSP was studied. It was indicated that two columns of opposite chirality gave reversed elution orders.
