A New Kinetic Method for the Determination of Laccase Activity Based on the Measure of the Intermediate

In reverse micelle dioctyl sulfosuccinate sodium salt (AOT)/ n octane, enzyme catalytic reaction of laccase o phenylenediamine system has super activity, and the intermediate is separated well from the product 2,3 diaminophenazine. The reactive calculation shows that there has linear relationship between the initiate rate of the intermediate and the activity of enzyme. The concentration of the intermediate was detected by using initiate rate method. A new kinetic method for the determination of laccase activity based on the measure of the intermediate was proposed. It has high sensitivity and selectivity based on the initial rate method. The linear range for laccase activity is 0~2.5U and the detection limit is 0.035 U.
