Determination of Total Flavonoids in Llex Kudincha by Three Wavelength Spectrophotometry

Determining the total flavonoids in Llex Kudincha by traditional spectrophotomeic method,baseline shift and asymmetric absorption peak were occurred on the absorption curve. The absorbance error of interfering components turbid solution ,Background changed with concentration changed and asymmetric absorption peak problems can be eliminated by three wavelength spectrophotomeic method. The flavonoids in Llex Kudincha with aluminate to produce stable complex which absorption there was red shift of longer wavelength . The regression equation of concentration ~Δ A was obtained:Δ A=-3.702×10 -3 +3.7023C ,correlation coefficient r =0.9994. The experimental results demostrate the total flavonoids concentrations in 600~1600 (mg/L) with Δ A obeying linear relation measured the absorbance at wavelength λ 1= 470 nm,λ 2=420 nm,λ 3=370 nm. The recovery is 98% ~112% and the coefficient of variation is 1.14%( n =9). The method is more advantageous than traditional spectrophotomeic method.
