Resonance Light Scattering Spectra of Cetylpyridinium Bromide and Deoxyribonucleic Acid System and Its Application to Deoxyribonucleic Acid Assay

A cationic surfactant, cetylpyridinium bromide (CPB) was used to determine deoxyribonucleic acids (DNA) at nanogram level with a resonance light scattering (RLS) technique. The electrostatic interactions between CPB and DNA gave rise to three characteristic peaks of RLS at 263 nm, 363 nm and 492 nm. Under optimal conditions, the enhanced intensity of RLS was proportional to the concentrations of DNA in the range of 0.2~2.0 mg/L for calf thymus DNA and 0.2~1.15 mg/L for fish sperm DNA. The corresponding limits of detection were 0.07 mg/L and 0.05 mg/L, respectively. The relative standard deviation of ten replicate measurements was 2.2% for 0.75 mg/L fish sperm DNA. This method was used to detect DNA in ~synthetic and real samples successfully.
