Determination of Trace Germanium in Ganoderma by Hydride Generation-Atomic Fluorescence Spectrometry

A method for the determination of total Ge in ganoderma samples by hydride generation-atomic fluorescence spectrometry (HG-AFS) was developed. By using microwave assisted digestion procedure, organic Ge could be destroyed successfully and element loss caused by the chlorine existing in samples could be avoided. Working conditions of AFS-920, optimum conditions for the generation of volatile of Ge species and microwave assisted procedure were optimized. Optimum acidity of the sample solution was 20% H_3PO_4 and the signal intensity was increased with the presence of 5% H_2SO_4. Comparison was made between the results obtained by two sample digestion procedures: microwave assisted procedure and dry ashing treatment. The detection limit and RSD of this method were 0.83 μg/L and 1.26%, respectively. The recoveries ranged from 84.0% to 96.5%.
