Determination of Plant Growth Regulater in wheat Seedlings by High Performance Liquid Chromatography

A high performance liqid chromatographic method was developed for determining the quantity of gibberellin(GA 3) and 3 indole acetic acid(IAA) in wheat seedlings .The wheat seedlings were homogenized in 80% methanol in a mixing vessel for 24hr at 4℃,filtered and the residue reextracted with 80% methanol for three times.The combined filtrate was evaporated to a small volume (20mL) under vacuum at 40℃(Water bath) and subsequently purified by forcing the solution through a PT C 18 cartridge with the aid of a 10mL syringe to remove the non polar compounds from the solution. The filtrate was then furtherreduced in volume (5mL) under vacuum at 40℃ and again forced through a PT C 18 cartridge .The filtrate was subsequently evaporated to dryness and taken up in 2mL of the mobile phase for reversed phase high performance liquid chromatography. It is essential that the H of the sample be adjusted to acidic.
