Separation, Purification and Character Research of Polysaccharide Extracted from Dioscorea Opposite Thunb

A water-soluble crude polysaccharide DT was obtained after extracting in optimal conditions, concentrating, precipitating with 4 times volume of absolute alcohol and desiccating. Two water-soluble polysaccharides named DTA and DTB were obtained by DEAE-Sepharose CL-6B chromatogra-phy. Purity detection indicated that DTA was homogeneous by gel filtration chromatography on Sephadex G-100 and cellose acetate pellicle electrophoresis. The monosaccharides of DTA were analyzed by using HPLC, the result showed that DTA was mainly composed of Fru and Glu, in the molar ratio of 1:26. 36.
