Comparison of Three DNA Fingerprint Analyzing Methods for Maize Cultivars' Identification

Three DNA fingerprint analyzing methods including distinct band method, primer combination method and core primer combination method had been generalized in this research and compared their usefulness in maize cultivars' identification. A set of 60 elite maize (Zea mays L.) inbred lines were assayed with 62 pairs of SSR primers. 238 alleles were found with average alleles 4.08,average polymorphism information content (PIC) 0.612 and average marker indices (MI) 2.58 per locus. The three index evaluating primer polymorphism (average alleles, PIC, MI) were not completely coherent. It was manifested from three methods' comparison that distinct band method was useful only in fixed materials. When the materials enlarged, samples originally with distinct band often no longer had distinct band; on the other hand, its efficiency was low due to need screening a lot of primers. Primer combination method could discriminate all materials by different primers' limited combinations, avoided of a lot of primer-screening. Core primer combination was an extension of primer combination method, data from different laboratories could be compared and integrated by using a set of fixed core primer combination and it would lead to standardization of maize DNA fingerprint analysis.
