Optimization of SRAP Reaction System for Plants of Dendrobium

Sequence-related amplified polymorphism (SRAP) is a new marker technique. In this paper, several significant parameters of SRAP reaction system were investigated and optimized by taking the total DNA of D.huoshanense as template. By abundant experiments, the stable reaction system was established: 25μl reaction system containing 30ng DNA template, 2.5mmol/L Mg2+, 0.8μmol/L forward primer, 0.8μmol/L reverse primer, 200μmol/L dNTPs and 1U Taq DNA polymerase. The SRAP reaction system is efficient to identify the species for the plants of Dendrobium and lays a foundation to study the genetic diversity research for Dendrobium.
