Developing Chromosome Segment Substitution Lines (CSSLs) in Cotton (Gossypium) Using Advanced Backcross and MAS

Chromosome segment substitution lines (CSSLs), also called as introgession lines (ILs), carry a sin- gle or fewer donor segments in the same background of a recurrent genotype. If only a single donor segment, they are called single segment substitution sines (CSSLs). In the study, a cotton population consisted of 116 CSSLs were developed from advanced backcrosses between the recipient, CCRI221, a cultivar of cotton, and donor hai1, a barbadense cotton by microsatellite marker-assisted selection (MAS). 116 BC4F1 lines were screened using 276 SSR primer pairs with polymorphism between ricipiend and donor in 25 linkage groups, which covered 2 347 cM, approximately 52.7% of cotton genome screened. There are 15 to 55 donor segments in lines with aver- age of 32.92, which cover 180.7 cM to 969 cM of cotton genome. Average size of cotton genome covered by substi- tution segments are 502 cM, covered 21.4% of cotton genome detected. Average donor segments are 0.1~8.3 in each linkage group of in each BC4F1 line, which average size covered each linkage group ranges from 0.2 cM to 54.2 cM, 10.3~35.5% of the average long screened, ranged from 0.5 cM to 207 cM, of each linkage group in every lines.
