Cloning and Genetic Characterization of the Fertility Restorer Gene Rf-D1(t) for Dian-Type Cytoplasmic Male Sterility of Rice (Oryza sativa L.)

Testing the seed-setting and pollen fertility of hybrid F1, Ansanbyeo and Nan34 performed high restoration ability to different cytoplasmic male sterile lines (CMS-DT1, CMS-BT and CMS-HL). According to the published rice genome sequence, primers were designed in the region of rice fertility restorer genes and a pair of molecular primers M43804 and M43558 which are closely linked to the restorer genes of two Dian-type CMS restorer lines Ansanbyeo and Nan34 was determined. Dian-type restorer gene Rf-D1(t) was firstly cloned with code of 16 PPR proteins. Sequence comparison showed that this gene has 99% similarity to BT-type restorer gene Rf-1. There was only one base pair difference between the two restorer genes in the section of ORF, i.e. the A of Rf-D1(t) changed into C of Rf-1 at 1 149 bp, and an amino acid code changing from K (lysine) to N (asparagines) was occurred. The difference is located in a PPR structure which means that amino acid might be one of the key loci of the restorer genes, and its alternation may affect the variation of restorability for rice CMS lines.
