Increase of Tocopherol Content in Endosperm by Overexpression of Homogentisate Phytyltransferase in Transgenic Rice

In order to increase the tocopherol content of rice, the homogentisate phytyltransferase (OsHPT) gene was transferred into an elite rice japonica cultivar Wuyujing3 driven with CaMV 35S promoter by Agrobacterium-mediated transformation RT-PCR analyses confirmed the expression of the input transgene in rice endosperm. HPLC results showed that after OsHPT was expressed in the rice, the total tocopherol content in endosperm of transgenic rice lines was significantly higher than the wild type, mainly for the increase in α-tocopherol content. In transgenic rice, the contents of total tocopherol and vitamin E increased maximally 2.5 and 1.8 folds respectively compared with those of wild type.
