Genetic Relationship Analysis of Apple Rootstock Resources Based on SSR Markers

Genomic DNA of 46 apple rootstock resources was analyzed using SSR method, which made us knowing the genetic background distribution of common stocks in the wild resources, and provided reference for the selection of the parents of stock breeding. In this study, 31 pairs of primers with high polymorphism were selected, and 111 polymorphic loci were obtained. The similarity coefficients of 46 apple rootstocks were between0.585 6~0.964 0, which were calculated by NTSYS software. Through constructing cluster tree by UPGMA method, 46 apple rootstock resources were divided into 4 groups: M. micromalus Makino was clustered alone as the first class; 7 varieties, Ser. Baccatae mainly were clustered together as the second class; As the third class,three varieties were included: M. sieboldii(Regel.) Rehd., M. rockii Schneid. and M. hupehensis(Pamp.) Rehd.;the fourth class mainly included Xinjiang local varieties, M. prunifolia varieties and several cultivated rootstock varieties, totally 32 varieties. The fourth class was clustered and divided into 4 groups, in the fourth groups most of the local varieties of Xinjiang and M. prunifolia clustered together, while cultivated rootstock varieties such as M9, Luo2, and Luo6 clustered with M. asiatica, M. sieversii and M. prattii(Hemsl.) Schneid. together.
