Committing E14 mouse embryonic stem cells to differentiate into thyrocyte-like cells in three-diamension collagen gel.

Objective To evaluate the feasibility of directing E14 mouse embryonic stem cells to differentiate into thyroid-like cells in vitro in three-dimension collagen gel without changing inducing factors.Methods E14 mouse embryonic stem cells were allowed to differentiate into embryoid bodies in suspension for 6 days and embryoid bodies were cultured in three-dimensional collagen gel for the next 11 days,with the addition of thyroid stimulating hormone and insulin from the beginning;On d14,KI was added until the end.Results Embryoid bodies grew well in Collagen Gel.The differentiated cells grew into the gel gradually.Electronic microscope showed that some induced cells were rich in Golgi's bodies and endoplasmic reticulums.A few secretory granules were also visible,which was similar to human adult thyrocyte.Conclusion E14 mouse embryonic stem cells can be differentiated into thyroid-like cells in collagen gel.Our study provided a new culture method capable of differentiating mouse embryonic stems cells into thyroid cells in vitro.
