EFFECTS OF IMMUNOREGULATORS ON NMDAR_1 AND NF-κB p65 EXPRESSIONS IN BRAIN OF RATS WITH PENTETRAZOLE-INDUCED EPILEPSY

Objective To explore the role of immunoregulator in epileptogenesis.Methods PTZ(pentylenetetrazol) was used to induce epilepsy,LMZ(levamisole) and DXM(dexamethasone) were used as immunoenhancer and immunodepressor respectively to interfere in the occurrence of epilepsy.The rats were randomly divided into 4 groups:NS(control)group,PTZ group,LMZ+PTZ group,and DXM+PTZ group.After treatment with relevant reagents,the behaviour was observed,the expressions of NMDAR1 and NF-κB in the cerebral cortex and hippocampus of rats were examined by means of immunohistochemistry.Results The behaviour observation indicated that no seizure happened in the control group,but Ⅲ-Ⅳ grade of seizure in the PTZ group and Ⅴ grade in the LMZ+PTZ group,and Ⅰ-Ⅱ grade in the DXM+PTZ group occured.Immunohistochemical staining showed that there were expressions of NMDAR1 and NF-κB in the cerebral cortex and hippocampus of all groups.The same tendency of changes for the two indices was observed.The expressions of NMDAR1 and NF-κB p65 in the PTZ group were higher than that in the NS group,and the expressions were further enhanced in the LMZ+PTZ group compared with those of the PTZ group,while they were lower in the DXM+PTZ group than in both PTZ and LMS+PTZ groups.There were significant differences in NMDAR1 and NF-κB immunoreactivities among the four groups.Conclusion Immunoenhancer or immunodepressor could up-regulate or down-regulate the expression of NMDAR1 and the activation of NF-κB could interfere the occurrence and development of epilepsy.
