Cloning and Sequencing of Metalloprotease Gene of A Pathogenic Vibrio Anguillarum

A pathogenic Vibrio anguillarum strain W 1 was originally isolated from diseased seabass ( Lateolabrax japonicus ) in China. Its extra cellular protease activity is 4226u/ml. The partially purified metalloprotease is toxic to turbot. The metalloprotease gene was cloned by PCR amplification from its chromosomal DNA. The nucleotide sequence was determined. The ORF of the gene consists of 1836 bp, which encodes a polypeptide of 611 amino acids. The nucleotide sequence is highly identical to that of a Vibrio anguillarum strain NB10. The similarities of the amino acid sequence of the metalloprotease with those of V. proteolyticus, V. vulnificus, V. cholerae, Aeromonas punctata, Aeromonas hydrophila are 73%, 70%, 69%, 53%, 51% respectively.
