Rapid and Quantitative Detection of Listeria monocytogenes Using Colloid Gold Immunochromatographic Method

Objective To develop a rapid and quantitative detection method on Listeria monocytogenes using colloid gold immunochromatographic method.Methods The rapid detection method was established with double-antibody sandwich assay,and the specificity and sensitivity of the method was then evaluated.Quantification was realized by constructing a standard curve,Milk powder,milk solution and ham sausage,added with ricn were tested to evaluate the feasibility on solid,semi-solid,liquid food samples and doubtful bioterrorism samples by this method.Results This established method could be accomplished qualitatively as well as semi-quantitative detection in 15 minutes,the sensitivity was up to 3.5×103 cfu/ml,the linear range was from 3.5×105cfu/ml to 3.5× 108cfu/ml,the recovery rate was 99%~101.7%.This method showed good specificity and sensitivity,which could detect samples directly.Conclusion The colloidal gold immunochromatographic test,which could detect the abrin in samples rapidly,specifically,sensitively and accurately,was able to realize quantification,and is suitable for field detection.
