The Potentiation of Inhibitory Effect of Pingyangmycin on Human Nasopharyngeal Carcinoma Cells by Coixenolide

FENG Hui qiang,QIU Jian xing,CHEN Ning, et al.//Guangdong Provincial Institute of Traditional Chinese Medicine,Guangzhou 510095
Objective:To Examine the feasible influence of coixenolide (CXL) on the antitumour effect of pingyangmycin (PYM,i.e.bleomycin A 5) on human nasopharyngeal carcinoma cell line CNE 2Z. Methods:Micro colony formation assay was used to test the sensitivity of CNE 2Z to PYM.Results:PYM inhibited the proliferation of CNE 2Z in a dose dependance manner, with an ID 50 =10 -5.976 ±10 -6.754 mol/L. Addition of CXL (10 -7 mol/L and 10 -6 mol/L) shifted both the dose effect curve and time response curve of PYM to the left, reducing the ID 50 of PYM to 10 -6.315 ±10 -7.056 mol/L and 10 -6.716 ±10 -7.420 mol/L,respectively,and shortening the duration of PYM's action CXL,however, did not potentiate the acute toxicity of PYM to normal mouse. Conclusion:CXL selectively enhances PYM cytotoxicity to CNE 2Z.
