Effects of dexmedetomidine on the expression of NF-κB-associated proteins in neonatal rats with cardiac inflammation induced by lipopolysaccharide

Objective To investigate the effect of dexmedetomidine on the expression of nuclear transcription factor-κB(NF-κB) signaling pathway in neonatal rat with cardiac inflammation induced by lipopolysaccharide(LPS). Methods SD neonatal rat cardiomyocytes were isolated and cultured and divided into control group, model group and test group. The model group and test group were treated with LPS(10 mg·L~(-1)) for 6 hours, while the control group were treated with equal amount of 0.9%NaCl. In addition, the test group was treated with dexmedetomidine(10 ng·mL~(-1)) for 1 hour before LPS treatment. The contents of nitric oxide(NO), tumor necrosis factor-α(TNF-α), prostaglandin E2(PGE2) in the supernatant of cultured cells were detected after 24 hours of treatment. The proteins associated with NF-κB signaling pathway were detected by Western blot. Results In the control group, model group and test group, the contents of TNF-α were(20.12±2.75),(55.35 ±1.57),(40.42±2.48) pg·mL~(-1), respectively; the contents of NO were(6.43±1.38),(35.69±1.48),(27.45±2.34)μmol·L~(-1),respectively; the contents of PEG2 were( 85. 26 ± 3. 44),( 346. 58 ± 14. 96),( 265. 46 ± 15. 98)pg·m L~(-1),respectively; the relative expression of NF-κB p65 protein were 0. 36 ± 0. 05,1. 89 ± 0. 12,0. 65 ± 0. 07,respectively; p-AKT were 0. 27 ± 0. 06,1. 52 ± 0. 24,0. 52 ± 0. 02 respectively. There were statistically significant differences between the model group and the control/test group( P 0. 05). Conclusion Dexmetiomidine pretreatment can inhibit the LPS-induced inflammation in neonatal rat cardiomyocytes and alleviate myocardial injury by regulating the expression of NF-κB pathway proteins.
