Effects of gliclazide on differentiation and Wnt signaling pathway of osteoblasts MC3T3-E1

Objective To investigate the effects of gliclazide on differentiation and Wnt signaling pathway of osteoblasts MC3 T3-E1. Methods Mouse osteoblasts MC3 T3-E1 cells were assigned to experimental-L/-M/-H group(1, 10, 20 μmol·L~(-1) gliclazide) and control group(0.9% NaCl). Cell counting kit-8(CCK-8) was used to detect the proliferation of MC3 T3-E1 cells, alkaline phosphatase(ALP) in MC3 T3-E1 cells and osteocalcin(OCN) in culture supernatant was detected, and the expression of Wnt signaling pathway protein in MC3 T3-E1 cells was detected by protein immunoblotting. Results After 24 hours of administration, the inhibition rates of MC3 T3-E1 cell proliferation in low, medium and high dose groups were(16.52±1.06)%,(26.85±2.46)%,(35.69±3.57)%, respectively. On Day 5 after gliclazide treatment, of the control group,in low, medium and high dose groups, the ALP content were(1.98±0.12),(2.15±0.09),( 2. 46 ±0. 15),( 3. 08 ±0. 14) U·g-1 prot,the content of OCN were( 18. 28 ± 1. 15),( 20. 24 ± 1. 12),( 22. 25 ± 1. 17),( 24. 25 ±1. 18) ng·m L-1. At 24 h after gliclazide treatment,the relative expression of Wnt1 protein were 0. 56 ± 0. 11,0. 63 ± 0. 05,0. 85 ± 0. 07,1. 02 ± 0. 14,the relative expression of low density lipoprotein related receptor protein( Lrp5) were 0. 25 ±0. 06,0. 45 ±0. 06,0. 68 ±0. 12,0. 89 ±0. 05 respectively,the relative expression of Lrp6 were0. 32 ± 0. 05,0. 49 ± 0. 11,0. 66 ± 0. 11,1. 05 ± 0. 16,the relative expression of β-catenin were 0. 29 ± 0. 06,0. 36 ± 0. 04,0. 49 ± 0. 02,0. 69 ± 0. 11. There were statistically significant differences between the control group and the experimental-L/-M/-H group( P 0. 05). Conclusion Gliclazide can promote the proliferation and differentiation of MC3 T3-E1 cells by up-regulating the expression of Wnt signal transduction pathway protein.
