Analysis of the genetic diversity of coconut (Cocos nucifera)by inter-simple sequence repeats (ISSR)technique

The genetic diversity of 30 coconut accessions from home and abroad was analyzed by using ISSR(inter simple se-quence repeats) technology.A total of 157 amplicons were generated with 29 primer pairs,which had higher polymorphism,and 110 amplicons were polymorphic bands.The genetic similarity ranged from 0.548 to 0.962,with the mean of 0.788,based on DICE software.The highest genetic similarity value was between Red dwarf(No.14)and hybrid(No.16),while the lowest one was between Hainan Tall(No.29) and Fiji Tall(No.5).A dendrogram was constructed by UPGMA method,and the ac-cessions were clustered into three groups(A,B and C) at a similarity coefficient criterion of 0.79.Group A was composed of all the Talls,Dwarfs and Hybrids,and all the special variants were clustered into Group B except for Xiongshu(No.22),Group C was consisted of all the accessions from Hainan province.The results suggest that coconut genetic diversity in China is not very high,and much more elite germplasm should be introduced.
