DNA Damage of Bone Marrow Cells in Mice Induced by Nano-Fe_3O_4 Particles

Objectives To observe the effect of Nano-Fe3O4 on the DNA damage of bone marrow cell in mice.Methods Forty clean grade ICR mice were divided into four groups by random with equal numbers of male and female in each group.They were high-dosage-treated group(40 mg/kg,1/4 LD50),middle-dosage-treated group(10 mg/kg,1/16 LD50),low-dosage-treated group(2.5 mg/kg,1/64 LD50),and pure water negative control group.Nano-Fe3O4 in a volume of 10 mL/kg and with a speed of 0.02 mL/s was injected once through tail vein.The DNA damage of bone marrow cells was detected three days after injection of Nano-Fe3O4 by using single cell gel electrophoresis.Results The percentage of DNA damage at grade 0 of all Nano-Fe3O4 treated groups were obviously lower than those in the control group(P0.05).There was a tendency of increasing the dosage of Nano-Fe3O4 with an obviously higher grade of DNA damage(P0.05).And the DNA damage at grade ≥Ⅱ was appeared only in the high-dosage-treated group.Conclusion Nano-Fe3O4 could cause DNA damage of marrow cells in mice injected intravenously after three days.
