Releasing of Tumor Necrosis Factor from Recombinant Escherichia Coli by Chemical Permeabilization and High Speed Bead Milling

The conditions of chemical permeabilization and high speed bead mill in disrupting recombinant E. coli to release TNF were investigated and optimized. Disruption time, guanidine hydrochloride concentration and pH of the solution dominating chemical permeabilization were investigated. Residence time, bead load, agitation speed and biomass concentration that affect high speed bead milling were studied. A comparison was made between the two methods.
