Auto-focusing function for microscope image based on weighted neighborhood correlation

A new auto-focusing function for microscope image based on weighted neighborhood correlation is presented,and the effect of pixel neighborhood grey level correlation and astigmatism in microscope image on the focusing results is analyzed based on the research of focusing principle and imaging processing.The 4-neighborhood grey level correlation of each pixel for every image in the focusing processing is computed.Then a quadratic polynomial focusing evaluation function, weighted average value based on the correlation,is calculated,and the weights are decided by the distance between the corresponding pixel and the image center.Finally,the image with the maximum function value is selected as the focusing image.The experimental results indicate that the focusing sensitivity factor increases by 0.318 5~0.326 8,and the focusing correct rate improves by 0%~40% under the noise conditions compared with the classical method.This method can accurately estimate the focusing extents with high sensitivity and strong noise immunity.
