PCR-RFLP Analysis on Edaphon Community Structure in Eucalyptus Plantation

In this research, we investigated the edaphon community structure from Gaofeng forest-farm Eucalyptus plantation in Guangxi Nanning by PCR-RFLP technique and 16S rDNA sequence analysis. Total DNA of microor- ganism in Eucalyptus plantation was extracted by using the direct method, then we constructed a 16S rDNA clone library of the soil microbes. A total of 192 positive clones were radomly chosen from the clone library to be examined. The PCR-RFLP result showed that 188 positive clones determinately contained 16S rDNA and they went back to 52 different operational taxonomic units (OTUs). We also radomly chose 35 clones among them to sequence and constructed phylogenetic development tree. The results indicated that the bacteria species in Eucalyptus plantation soil are relatively abundance. Further sequence analysis showed that the dominant bacteria in this Eucalyptus plantation soil were Acidobacteria and Proteobacteria which was 40% and 27%, respectively. This resarech investigated the edaphon community structure of Eucalyptus plantation, which would provide the foundation information for studying the variety in environmental quality of Eucalyptus plantation.
