EFFECT OF REACTIVE OXYGEN SPECIES ON ERYTHROCYTE MEMBRANE FLUIDITY

Objective:To study the mechanism of erythrocyte oxidative hemolysis caused by reactive oxygen species (ROS) Methods:Different concentrations of H 2O 2 were acting on vitro normal erythrocyte,and the erythrocyte hemolysis degree,erythrocyte membrane lipid peroxide (LPO) and erythrocyte membrane microviscosity (EMMV) were determined in colorimetry method, thiobarbituric acid fluorescence method , and fluorescence polarization method, respectively Results:The erythrocyte oxidative hemolysis happened when erythrocyte were acted upon by H 2O 2 with 50mmol·L -1 , 100mmol·L -1 ,120 mmol·L -1 , the erythrocyte hemolysis degree, erythrocyte membrane LPO and EMMV were increased The effects of the increasing erythrocyte hemolysis degree, erythrocyte membrane LPO and EMMV were obviously dose dependent within extent of 50 mmol·L -1 ~120 mmol·L -1 H 2O 2 There was significant positive correlation between erythrocyte hemolysis degree and EMMV There was also significant positive correlation between erythrocyte membrane LPO and EMMV Conclusion:A part of mechanism of erythrocyte oxidative hemolysis caused by ROS leads to erythrocyte membrane lipid peroxidation, and decrease of erythrocyte membrane fluidity is induced by erythrocyte membrane lipid peroxidation
