Production of Xylanase by Aspergillus niger A3 Using Solid State Fermentation

A high xylanase producing fungus Aspergillus niger strain A3 was isolated and its fermentation conditions by solid state fermentation were studied. The optimal culture conditions were as follows, initial pH 4.6, temperature 28℃, 1ml spore suspension inoculum, wheat brantbagasse = 1:1.5 and cultivations days. The xylanase activity was as high as 5147 IU/g dry medium. Nitrogen source Component, temperature, pH and cultivation time had great effects on the ratio of xylanse and cellulase. Some properties of the xylanase produced in liquid and solid cultures were compared. Both the optimal temperature were 55℃, optimal pH were 4 .6 and 4 .2 respectively. Xylanase synthetized in solid fermentation was little more themostatble than that from liquid cullture and the temperature for loss of half activity (t 1/2)in 1 h were 54℃ and 51℃ respectively.
