Effect of N-acetylcysteine on Oxidation and Antioxidation in Rats with Acute and Chronic Smoking

Objective: To assess the effect of N-acetylcysteine (NAC) on oxidation and antioxidation in rats with acute and chronic smoking. Methods: Wistar rats were randomly divided into three groups: control group (6), smoking group(30), intervention group(smoking+ NAC, 30). The latter two groups were subdivided respectively into five different periods groups: (1, 2, 7, 14 and 60 d). Thiobarbituric acid method, Ellman method and Rad1oimmunoassay were used respectively to detect the lipid peroxides (LPO), glutathione (GSH ) and superoxide dismutase(SOD). Results: It was shown that, in the acute and chronic smoking rats,serum LPO significantly increased; but GSH in whole blood and bronchial alveolar lavage fluid (BALF)and SOD in serum and BALF significantly decreased when compared with the control groups (P 0. 05). After the intervention with NAC, GSH and SOD significantly increased, while LPO decreased when compared with the smoking groups (P<0.05). Conclusions: NAC, through exerting its direct or indirect antioxidation and inactivation function against free radical induced by smoking, may play a important role in the prevention and treatment of COPD.
