New Method for the Preparation of Indole-3-Pyruvic Acid

Indole-3-pyruvic acid, an important intermediate in the preparation of tryptophan by the method of "Chemical-Enzyme", was prepared by a new method from indole in this paper. Indole-3-pyruvic acid was synthesized sequentially through formylation, condensation and hydrolysis reactions, and the major conditions for the reactions were optimized. Using triethylamine as catalyst and water as solvent, the yield of condensation product could reach 81. 5% if the optimum reaction conditions of time 2. 5 h, temperature 110℃ and substrate concentration 2 mol ·L-1 were selected. When protected by N2 , and selected component of alkali 5: 1, temperature 110℃ , substrate concentration 0. 5 mol·L-1 and time 2 h, the productivity of the hydrolysis reaction could remain 92%. All these datas gave the possibility to the scaling-up for the production of indole-3-pyruvic acid.
