Separation and Identification of an Acetic Acid Bacterium and Its Mutation Breeding by Ion Implantation

An acetic acid bacterium S12 was isolated from the solid fermentation substrate of Shanxi overmature vinegar.It was identified as Acetobacter pasteurianus.SM12-87 was obtained from the strain S12 by N+ implantation.The conversion rate of alcohol to acetic acid was 88.98%;the yield of total acid and ethyl acetate was 91.67 g·L-1 and 1.143 g·L-1 respectively under 10%(V/V) ethanol,quiescently cultured 5 d at 30℃ by SM12-87.The liquid state seed of SM12-87 was inoculated into the solid fermentation substrate of Shanxi overmature vinegar.The result showed that the yield of acetic acid and ethyl acetate had increased by 20.57% and 41.83% respectively.So,this is a feasible way to optimize the microbial community structure and improve the yield and quality by mutation breeding of the wild functional strain separated from traditional brewing process.
