Genetic Transformation of Foxtail Millet Mediated by Agrobacterium Tumefaciens

Wang Yongfang Li Wei Diao Xianmin (Institute of Millet Crops, Hebei Academy of Agriculture and Forestry Sciences, Shijiazhuang 050031)  
Factors influencing the efficiency of genetic transformation of foxtail millet via Agrobacterium tumefaciens co-culture were studied and a suitable procedure was established in this paper. Callus derived from immature inflorescences were the best adapter for initiating co-culture. The suitable stage of Agrobacterium for co-culture was when its growing concentration reaches OD600 0. 5 and 100 μM of AS added to the co-culture media gave the highest rate of transformation. Lower co-culture temperature, such as 22℃ other than 28℃ , show a higher transformation efficiency. The procedure of transgenic resistant callus selection and plantlet regeneration were also reported in this paper. Using the system described here a few cultivars of foxtail millet such as Yugu 1, Gao 39 and Lugu 10, were transformed via Agrobacterium strain LBA4404 and EHA101, so far 35 transgenic plantlets were obtained.
