Polymorphism Analysis by Sequence-related Amplified Polymorphism Markers in Brassica napus L

Polymorphism among 1141B,KenC_1,32B and 32C were analyzed with SRAP(sequence-related amplified polymorphism) markers.There were 13-36 clear polymorphic combination bands per primer,and the average was 5.8.About 419 amplified fragments and 116 polymorphic fragments were detected 20 SRAP primer combinations.The percentage of polymorphic fragments was 28.23%.The number of amplified fragments and polymorphic fragments per primer combination were 20.95 and 5.8,respectively.The polymorphism analyzed by F_(2) individual of 1141B Ken C_1 with 24 primer combinations showed that there were 75 polymorphic bands,the average was 3.12.The χ~2 test for allelic genotype of all loci indicated that 1.33%of the SRAP markers distorted from the expectation 3∶1 ratio(dominance markers) and 1∶2∶1(co-dominance markers).It seems that SRAP markers was a high efficiency marker system in the paper.It showed that SRAP were an effective,cheap and dependable marker for polymorphism analysis and linkage map construction in B.napus L.
