The Effect of Lycopene on the Differentiation of Osteoclast Induced from Bone Marrow in Mice

The effect of lycopene on the differentiation of osteoclast was induced from bone marrow in vitro.Mononuclear cells of mice bone marrow were incubated with DMEM containing macrophage colony stimulating factor(MCSF) and receptor activator of NFκB ligand(RANKL).The culture was treated with lycopene of different concentrations(10-8,10-7,10-6mol/L).On the 7 d,the culture cells were fixed and were stained for tartate-resistant acid phossphatase(TRAP).The formation of osteoclasts was quantified by counting the number of TRAP+ multinuclear cells.The percentage of bone resorbed surface,TRAP activity and the expression of receptor activator of NFκB(RANK) mRNA were analyzed.The appearance of reduced mice osteoclasts was typical.Compared with the control group,the number of TRAP+ multinuclear cells,TRAP activity and the percentage of bone surface in lycopene middle-dose group and lycopene high-dose group significantly decreased(p0.05).But the expression of RANK mRNA significantly increased(p0.05)by being treated in different dose of lycopene.Lycopene can inhibit osteoclasts differentiation in vitro.
